Recent Trends in Chemistry and Pharmacology-Based Research Using Pyrrolidine Scaffolds: An Up-to-Date
Indian Journal of Pharmaceutical Education and Research
Abstract
Pyrrolidine is a saturated five-membered nitrogen-containing heterocycle widely recognized as a privileged scaffold in medicinal chemistry. Its conformational flexibility, strong basicity, and hydrogen-bonding capability contribute to its broad pharmacological relevance. Pyrrolidine derivatives exhibit diverse biological activities, including antimicrobial, anticancer, antiviral, anti-inflammatory, neuropharmacological, and antidiabetic effects. Several clinically approved drugs such as Paroxetine, Vincristine, Remdesivir, and Saxagliptin highlight its therapeutic versatility. Advances in synthetic strategies, including intramolecular cyclization, reductive amination, hydroamination, and lactam reduction, have expanded access to structurally diverse derivatives. This review summarizes structural features, synthetic methodologies, pharmacological applications, and emerging research directions of pyrrolidine scaffolds in modern drug discovery. Although they possess immense potential, challenges such as stereochemical complexity, possible toxicity, and metabolic instability remain critical factors in their development. Looking ahead, the integration of green chemistry, stereoselective synthesis, and computational drug design is expected to unlock new opportunities, making pyrrolidine scaffolds vital for next-generation pharmaceuticals and multifunctional materials. This review provides an updated overview of the structural features, synthetic strategies, pharmacological applications, and future perspectives of pyrrolidine-based research, highlighting its significance in advancing both chemistry and biomedical sciences.
